Histology and Diagnostic Core

The objective of the Histology and Diagnostic Core is to provide histopathology support, both technical and interpretative, for all projects. For Project 1, we will process congenic murine tissues for light microscopic analysis, immunostaining and electron microscopy. Scoring of murine kidneys for disease severity will be performed blindly. For Project 2, laser capture microdissection of renal tissue compartments will be performed on human kidney biopsies for quasispecies analysis. In addition, transgenic mice expressing vpr will be studied by light microscopy, electron microscopy and immunohistochemistry. For Projects 3 and 4, kidney tissue from podocan knockout and overexpressing mice, Sdk-1 overexpressing mice, Tg 26 mice crossed with Sdk-1 knockout mice, Tg26 mice crossed with TLR7 and TLR8 knockout mice, and Tg26 mice crossed with Stat3 mutant mice will be studied by light microscopy, electron microscopy, and immunohistochemistry. To validate the relevance to human disease in projects 1, 3 and 4, immunohistochemistry will be performed on archived human kidney biopsies for proteins of interest.
