Research

The primary focus of Dr. Klotman's laboratory is the molecular pathogenesis and therapy of Human Immunodeficiency Virus 1 (HIV-1) infection. Ongoing molecular pathogenesis studies include establishing the role of virus and host factors in determining disease progression in HIV-1 infection. One of the project studies the role of viral proteins in HIV-associated nephropathy as part of a multi-project NIH-funded initiative to understand the cell biology, virology and genetically determined host factors involved in this devastating manifestation of HIV. Recent important observations made by this group include the demonstration that HIV resides in and evolves separately in kidney cells and that anti-retroviral therapy can relieve some of the clinical manifestations related to HIV-associated nephropathy but the virus persists at that site despite therapy. Additional projects include elucidation of the relationship between innate immune mediators and HIV, particularly as part of the complex interaction between other sexually transmitted diseases and HIV. Ongoing work on this project includes the isolation and characterization of active HIV-1 inhibitory protein(s) derived from CD8+ cells and determining the role and mechanism of action of host antimicrobial peptides and toll-like receptor activation in suppressing and/or inducing HIV. These projects have both a basic science component focused on understanding the complex interactions between HIV and the vaginal and cervical mucosa that facilitate viral entry in the setting of other sexually transmitted diseases as well as a focus on understanding local immune responses that might be altered by candidate topical microbicides under development to prevent transmission.
